The train is by far the best way - cheap, easy and *much* more comfortable than the bus. The distance by rail between the cities is 190 km.
I don't know the train fare, but it's probably somewehere between US$10 and $20.

Train timetable Odessa - Chisinau:
Train 642 goes daily from Odessa Glavnaja 11:40 to Chisinau 16:46.
Train 236 goes on even dates from Odessa Glavnaja 12:51 to Chisinau 17:06.
Train 658 goes daily from Odessa Glavnaja 17:57 to Chisinau 22:36.
